Is ttyS0 on a header while ttyS1 is on the card (on an AT machine)? 
Perhaps your header cable is defective.  There are two wiring patterns
(without showing you exactly how the card's pins are mapped to DB25
pins)

If you see what I mean.  Using the wrong pattern is sure to stop the
port from working!  (did you get this stuff from a junk box?)
