No. It uses the milter interface of Sendmail, and thus absolutely
requires Sendmail.
When I migrated work off Sendmail to Postfix, I started using 
amavisd-new and I've been fairly happy with that. With Postfix you don't
have to use it as an outward-facing SMTP proxy, Postfix supports using
it as a content filter over LMTP.
Amavisd-new seems a *lot* more efficient than MimeDefang, too.
