Not easily. Debian (like most distros) makes a lot of assumptions about 
the root FS being writeable, though I gather that's being improved. It'd 
be a LOT more work than using knoppix - remember, Knoppix is at heart a 
cd-based debian system.

As others have suggested, modifying Knoppix is probably your best bet.
